Oxidation of trp to kynurenine
WebTDO and IDOs are heme-containing enzymes that catalyze the oxidation of L-Trp to N-formyl-L-kynurenine. This is the first rate-limiting step of the Trp-KYN system, which regulates the systemic level of Trp in the body. IDO isoform 1 (IDO1) also catalyzes a stereoisomer D-Trp which is a product of the gut microbiome.

WebAug 21, 2024 · An increase in plasma [Kyn] can occur when Trp oxidation exceeds renal handling, eg, after robust induction of TDO or IDO or loading with Trp or Kyn. Measurement of plasma Kyn metabolites could be more informative. WebSep 17, 2024 · The aryl hydrocarbon receptor (AhR) is a nuclear protein which, upon association with certain endogenous and exogenous ligands, translocates into the nucleus, binds DNA and regulates gene expression. Tryptophan (Trp) metabolites are one of the most important endogenous AhR ligands. The intestinal microbiota is a critical player in human …
WebTryptophan (TRP) metabolism and disposition are reviewed with particular emphasis on the hepatic kynurenine pathway (HKP) in health and disease. Over 95 % of dietary TRP is metabolized in the HKP, which is controlled mainly by tryptophan 2,3-dioxygenase (TDO) and produces important metabolites affecting functions in the brain and periphery. WebDec 15, 2004 · Bicarbonate-mediated SOD1-peroxidase-dependent oxidation of tryptophan to kynurenine-type products; MALDI-TOF analysis. (A) hSOD (1 mg/ml) was incubated with H 2 O 2 (1 mM) and NaHCO 3 (25 mM) in phosphate buffer (0.1 M, pH 7.4) containing DTPA (0.1 mM) for 1 h at room temperature. (B) Same as above but without HCO 3−.
